Cell Wall Synthesis

Creating cell walls of bacteria

Cell Wall Synthesis

Mechanism:

Fundamentals:

  • Intracellular pressure in cells is enormus
    • Cells walls are bolstered by Peptidoglycan to stabilize the cell
  • Three Stages:
    • Synthesis of Precursors
    • Synthesis of Linear Peptidoglycal Strands
    • Crosslinking

Synthesis of Precursors:

  • NAG-Uridine Diphosphate -(Phosphoenolpyruvate (PEP) and Enzyme)-> NAM-UDP
    • Isomerism of NAG-UPD to NAM-UDP w/ help of PEP and enzyme
Prevention Through Fosfomycin
  • NAM-UDP -(ATP and Enzymes)-> NAM-UDP-(AA)-AELAA
    • Creation of the Park Nucleotide
      • AA sequence attached to NAM-UDP: L-ala-D-glu-L-lys-D-ala-D-ala
Prevention Through Penicillin
Prevention Through Cycloserine
Prevention Through Vancomycin
  • (AA)-NAM-UDP --> (AA)-NAM-DP-C55Terp
    • Lipid I creation: exchanging Uridine for C55Terp - Membrane Bound Carrier
  • (AA)-NAM-DP-C55Terp --> NAG-(AA)-NAM-DP-C55Terp
Prevention Through Moenomycin
    • Lipid II creation: attaching NAG from the Lipid I
  • Transported out of the membrane
  • Creates the core strands of the peptidoglycan
